Dosage

Premedication: Oral dexamethasone 20 mg approx 12 & 6 hr before paclitaxel inj or diphenhydramine (or its equiv) 50 mg IV 30-60 min prior to paclitaxel inj & cimetidine (300 mg) or ranitidine (60 mg) IV 30-60 min before paclitaxel inj. Metastatic carcinoma of the ovary or breast who have failed standard therapy 175 mg/m2 IV over 3 hr every 3 wk. Adjuvant treatment of node +ve breast cancer 175 mg/m2 IV over 3 hr every 3 wk for 4 courses administered sequentially to doxorubicin-containing combination chemotherapy. After failure of initial chemotherapy for metastatic disease or relapse w/in 6 mth of adjuvant chemotherapy 175 mg/m2 IV over 3 hr every 3 wk. Myelosuppressive regimen 135 mg/m2 IV over 24 hr followed by platinum compd w/ a wk interval between courses. Non-small cell lung carcinoma 135 mg/m2 IV followed by cisplatin 75 mg/m2 every 3 wk. AIDS-related Kaposi's sarcoma 135 mg/m2 IV over 3 hr every 3 wk or 100 mg/m2 IV over 3 hr every 2 wk.

Indication

Combination w/ cisplatin: 1st-line & subsequent therapy of advanced carcinoma of the ovary & 1st-line treatment of non-small cell lung cancer. Adjuvant treatment of node +ve breast cancer sequentially administered w/ doxorubicin-containing chemotherapy. Breast cancer after failure of combination chemotherapy for metastatic disease or relapse w/in 6 mth of adjuvant chemotherapy. 2nd-line treatment of AIDS-related Kaposi's sarcoma.
